The CAARE project was represented at the NVMS Spring/Summer Meeting in Leiden, the Netherlands, where Carley A. Hinrichs, a doctoral candidate at Leiden University Medical Center (LUMC), presented her latest research on advanced analytical methods for AAV characterization.
Organized by the Dutch Society for Mass Spectrometry (NVMS), the meeting brought together researchers from academia and industry to discuss emerging developments in mass spectrometry and its broad range of applications in the life sciences.
In her poster, Nano-Reverse Phase Liquid Chromatography-Mass Spectrometry Methods for In-Depth AAV Capsid Characterization, Carley introduced an ultra-sensitive nano-RPLC-MS workflow designed to analyze adeno-associated virus (AAV) capsids using exceptionally small sample volumes. The method enables researchers to separate and quantify viral protein species, assess VP1/VP2/VP3 stoichiometry, identify truncated proteins, and detect post-translational modifications such as acetylation and phosphorylation.
These capabilities provide valuable insights into capsid composition and heterogeneity—important parameters for evaluating the quality, stability and consistency of AAV-based products. The workflow also offers potential for batch comparison studies and monitoring throughout downstream processing, while requiring only minimal amounts of material.
As gene therapies continue to evolve, robust analytical approaches are becoming increasingly important for understanding and controlling complex biological products. Carley’s work demonstrates how highly sensitive mass spectrometry can contribute to this effort by delivering detailed molecular information from even the smallest samples.
